I27. the Oxidation of Pyruvate by Heart Muscle
نویسنده
چکیده
BRAuN-MENENDEZ et al. [1939], working with heart-lung and isolated heart preparations, have shown that pyruvate can be utilized by the mammalian heart. Minced heart muscle, as will be seen later, oxidizes pyruvate at about the same rate as the intact heart, and it was used in the present work for a study of the mechanism of the oxidation of pyruvate. The object of the investigation was to determine whether oxidation occurs by way of the citric acid cycle as in pigeon breast muscle [Krebs & Eggleston, 1940]. The main evidence for the occurrence of the citric acid cycle in pigeon breast muscle is as follows: (1) The oxidation of pyruvate is inhibited by malonate. (2) The malonate inhibition is partially overcome by the addition offumarate, and the extra amount of pyruvate used in these conditions is in the proportion of 1 mol. of pyruvate to each mol. of added fumarate. In the presence of malonate succinate is formed in amounts equivalent to the fumarate added, the reaction being fumarate +pyruvate + 202 succinate + 3CO2+ H20. (1)

Inhibition by pyruvate of pig heart mitochondrial glutamate influx.
Cardiac muscle mainly oxidises fatty acids, ketone bodies and pyruvate originating from either glycolysis or glucogenolysis, or from lactate oxidation. The formation of pyruvate and its oxidation by mitochondria are controlled directly by the reoxidation of extra-mitochondrial NADH, which cannot operate directly via the respiratory chain [ 1,2] . This reoxidation probably occurs via the Borst c...
متن کاملEffects of Dichioroacetate on the Metabolism of Glucose, Pyruvate, Acetate, 3- Hydroxybutyrate and Palmitate in Rat Diaphragm and Heart Muscle in vitro and on Extraction of Glucose, Lactate, Pyruvate and Free Fatty Acids by Dog Heart in vivo
1. The extractions of glucose, lactate, pyruvate and free fatty acids by dog heart in vivo were calculated from measurements of their arterial and coronary sinus blood concentration. Elevation of plasma free fatty acid concentrations by infusion of intralipid and heparin resulted in increased extraction of free fatty acids and diminished extractions of glucose, lactate and pyruvate by the heart...
متن کاملEffects of re-feeding after prolonged starvation on pyruvate dehydrogenase activities in heart, diaphragm and selected skeletal muscles of the rat.
We investigated the capacity for pyruvate oxidation in skeletal muscle, diaphragm and heart after starvation and re-feeding. Starvation for 48 h decreased pyruvate dehydrogenase (PDH) activity in soleus (by 47%), extensor digitorum longus (64%), gastrocnemius (86%), diaphragm (87%), adductor longus (90%), tibialis anterior (92%) and heart (99%). Chow re-feeding increased PDH activity in all mus...

متن کاملThiamine and Cocarboxylase Concentration
In patients with congestive heart failure, thiamine deficiency has been demonstrated by the test-dose method (1, 2), by measurement of the lactate-pyruvate ratio of the blood (3), and by determination of blood cocarboxylase (4). The oxidation of pyruvate to acetate requires thiamine in the form of cocarboxylase (thiamine pyrophosphate). Olson, Pearson, Miller, and Stare have shown that thiamine...
